A new potent agent for neurogenesis in the hypocampus combination.

The present invention relates to a new potent and valuable neurotrophic composition which stimulates human-derived neural stem cells in the hippocampus to form new neurons in vitro and stimulates neurogenesis in mouse hippocampus under the same pathways in vivo. 
It also acts on human brain undifferentiated neurons stem-cells (hNSCs) causing their proliferation, a rare finding on nerve growth factors studys. 
It has no or low toxicity, another advantage is once a day protocol, taken at night, before going to bed. Little to no side-effects.
The present combination has the potential use of treating depression and cognitive impairment.
The invention also shows to exceed Prozac(fluoxetine) neurotrophic proprieties in the hypothalamus on mice and non-human primates to an ample extend.
It is safe such that does not trigger the benign proliferation of astrocytes, which already occurs with age and it is considered a maladaption.

BUSPIRONE:

Buspirone is a drug from the aryl(guetaryl)piperazines  that contains an active 1-(2-pyrimidinyl)-piperazine metabolite .

Buspirone processes partial agonist activity at 5HT-A1 receptors and a potent one at presynaptic 5HT-A1 autoreceptors. Which makes it a SRA(Serotonin releasing agent) after desensitization of the autoreceptors.

Buspirone also blocks inhibitory presynaptic D2 autoreceptors while the piperazine moiety inhibits the a2-andrenoreceptors, only at higher doses buspirone antagonizes postsynaptic D2 receptors. 
What this means is that a low dose of buspirone acts as a stimulant it increases noradrenergic and dopaminergic activity, while higher doses start to behave more like an typical anti-psychotic.

Various study's have shown this; For example if you take a sample of humans and let them take methamphetamine after a dose of buspirone, the subjective high of the drug is increased.
Animals (non-human primates) on low buspirone doses administer cocaine and amphetamines for longer periods of time and increasing buspirone dosage makes them suddenly stop self-administer the narcotics, since dopamine receptors are being blocked.

This drug is forgotten and listed only as a augmentative anti-depressant with poor results, it was never though of a stimulant.

Buspirone co-administer on different embodiment's, not only acts as a stimulant, but it has similar high to cocaine if crushed and insuflatted.

MELATONIN:
Melatonin (N-acetyl-5-methoxytryptamine, MLT) discovered in 1958 function as a neuroendocrine hormone in humans and it is the most potent antioxidant ever found, even after decades of synthetic or not antioxidants being introduced or discovered.
If you ask any person in anti-aging field they would say that melatonin is for sleep and circadian rhythms, has some anti-oxidant power and they will stop around there. The general public perception is that it helps to sleep.
